No the 200 OK should not contain Route header fields from the INVITE request. All being well these Route headers should have been stripped out by intermediate proxies before reaching the UAS anyway. Route headers are only useful for routing requests, therefore are not needed for responses.
 
The reason for including the Route headers from the INVITE in the CANCEL request is to ensure that the CANCEL takes the same route through the SIP network as the INVITE it is cancelling.

As per RFC 3261 "If the request being cancelled contains a Route header field, the CANCEL request MUST include that Route header field's values". 
 
Should 200 OK sent for the Cancel,  also contain such values. If so wat is the need of it?? 
 
Aren't only mandatory fields enough???
