Hi Bhanu,
     For testing the call transfer you need atleast 3 SIP Softphones to run your system. Few softphones like SJ Phones, X-Lite  Phone and Zoiper phone are freely available. And they allow you to change the SIP ports. Just configure the different SIP ports on each phone and run the instance of those phones. e.g.Configure SIP port of one phone on 5060, second phone on 5070 and third phone phone on 5080 and run the phones.By this way you can test the scenario’s like call transfer, call waiting, call hold when third party call will be received etc.

I am testing Class five features like Call transfer etc.....It will be helpful for me in testing if all 3 instances of Sofphone are running in same system to analyze & work on it.

Can you explain me your exact requirement? Because instead of using multiple instances of same Softphone you can use two different softphones which supports SIP. But you should make sure that port assigned for both the UA are different. Or else you will face port binding problem.

Even if softphone provides manual configuration of SIP listening port. Your softphone should be capable of running multiple instances of the application. And I guess X-Lite does not support multiple instances. 
 
Others pls correct me about X-Lite behavior, if I am wrong.

I am using Xlite Softphone,i tried for option to change the Port of Softphone but i dodn't find that option .I have noticed only the Port option for Proxy server.Please let me know the steps or where can i change the Host port of Xlite.
